I have created a new Temp tablespace (locally managed temp), and have been wondering whether it would adversely affect application users, if they are connected and active, if I change their default temp tablespace, to be directed to the new one.

Just go for it, change their temporary tablespace, no matter if they are connected or not, active or not. If some session is performing disk sort while you change their user settings, the current sort will continue in the old temp tablespace. Once that sort finishes, all the subsequent sorts will use the new temp tablespace. Users will not not notice the change at all.
